Nitrosigine® (Nutrition 21) is a patented, inositol-stabilized arginine silicate complex specifically engineered to overcome the poor bioavailability of standard L-arginine. With over 30 studies supporting its efficacy, Nitrosigine significantly elevates and sustains plasma arginine and nitric oxide levels for up to 6 hours — making it one of the most researched and effective pump and performance ingredients in the pre-workout market.

Studied Dose 1,500 mg/day; most studies use 1,500 mg taken 30–60 minutes pre-exercise
Active Compound Inositol-stabilized arginine silicate — Nitrosigine® by Nutrition 21 (patented complex)

Superior nitric oxide and arginine elevation

Nitrosigine produces significantly greater and longer-lasting increases in plasma arginine and nitric oxide vs. standard L-arginine. The inositol-silicate complex stabilizes arginine, increases intestinal absorption, and extends the duration of NO elevation to 6 hours — vs. L-arginine's 1–2 hour window.

Blood flow and muscle pump

Elevated nitric oxide dilates blood vessels via the arginine-eNOS-NO pathway, increasing muscle blood flow, nutrient delivery, and the pump sensation during resistance training. Clinical studies confirm significantly greater brachial artery flow-mediated dilation vs. L-arginine and placebo.

Cognitive performance and focus

A 2020 clinical study found a single dose of Nitrosigine significantly improved cognitive flexibility and executive function in active young men vs. placebo — effects attributed to increased cerebral blood flow via nitric oxide-mediated vasodilation. Benefits were measurable within 15 minutes of ingestion.

Recovery and muscle damage reduction

Nitrosigine reduces creatine kinase and other muscle damage biomarkers post-exercise, suggesting a protective effect on muscle tissue during intense training. Faster recovery between sessions supports greater training frequency and progressive overload.

1

eNOS-mediated nitric oxide production

Arginine (released from the Nitrosigine complex) is the exclusive substrate for endothelial nitric oxide synthase (eNOS), which converts arginine to citrulline and NO in vascular endothelial cells. NO activates soluble guanylate cyclase, increasing cGMP and triggering vascular smooth muscle relaxation and vasodilation.

2

Silicon bioavailability contribution

The silicate component of Nitrosigine contributes orthobiological silicon to the complex — a mineral that supports vascular endothelial integrity, collagen synthesis in vessel walls, and endothelial function. Silicon may contribute to endothelial health effects beyond the direct arginine-NO pathway.

3

Inositol-mediated cellular signaling

Inositol in the Nitrosigine complex contributes phosphatidylinositol signaling pathway activity, which plays a role in insulin signaling, cellular membrane function, and potentially in the enhanced arginine absorption and retention observed vs. standard L-arginine.

Nitrosigine vs. L-Arginine and L-Citrulline for Blood Flow — RCT
PubMed

RCT comparing Nitrosigine (1,500 mg), L-arginine (1,500 mg), L-citrulline (1,500 mg), and placebo on plasma arginine levels and blood flow markers in healthy men.

Healthy men. Acute crossover pharmacokinetic and vasodilation study.

Nitrosigine produced significantly higher and longer-lasting plasma arginine elevations vs. L-arginine and L-citrulline at equal doses. Greater flow-mediated dilation observed with Nitrosigine. Arginine levels remained elevated for 6 hours vs. 1–2 hours for L-arginine.

2
Nitrosigine and Cognitive Performance — RCT
PubMed

RCT of Nitrosigine (1,500 mg) vs. placebo measuring cognitive flexibility and executive function in active young men.

Active young men. Acute cognitive assessment study.

A single dose of Nitrosigine significantly improved cognitive flexibility scores vs. placebo within 15 minutes. Executive function and processing speed also improved. Consistent with improved cerebral blood flow via NO elevation.

Common Potential side effects

Very well tolerated across all published human studies
No significant adverse effects reported at 1,500 mg/day clinical dose
Mild GI discomfort possible in sensitive individuals — take with food

Important Drug interactions

Erectile dysfunction medications (sildenafil, tadalafil) — both increase cGMP/NO; additive vasodilation risk; avoid combining
Antihypertensive medications — additive blood pressure-lowering effects; monitor
Anticoagulants — arginine may mildly affect platelet function; monitor with warfarin
